Description

Liraglutide is a long-acting glucagon-like peptide-1 (GLP-1) receptor agonist. It is a sterile, white to off-white lyophilized powder used as the Active Pharmaceutical Ingredient (API) for the manufacture of injectable solutions. Chemically, it is an acylated human GLP-1 analog with 97% sequence homology to endogenous human GLP-1(7-37). The molecule is modified by the attachment of a C-16 fatty acid chain via a glutamic acid spacer at lysine residue 26. This structural modification allows liraglutide to bind reversibly to albumin in the blood, thereby protecting it from rapid degradation by dipeptidyl peptidase-4 (DPP-4) and slowing its renal clearance. This results in a prolonged half-life of approximately 13 hours, enabling once-daily subcutaneous administration.
